混合脐带血浆对脐血单个核细胞增殖及凋亡的影响

 　【摘要】 目的 通过检测混合脐带血浆（CBP）对脐血单个核细胞增殖及凋亡的作用，探讨其刺激造血细胞增殖的作用机制。方法 采用5′-溴脱氧尿嘧啶核苷（BrdU）掺入法检测CBP对脐血单个核细胞DNA合成的影响;Western Blot检测CBP对细胞周期素cyclin A、cyclin D 1 及细胞周期素依赖性激酶CDK2和CDK4表达的影响;姬姆萨染色形态学观察和流式细胞术PI染色法检测CBP对细胞凋亡的作用。结果 （1）加入10%CBP12h后BrdU检测值与对照组（10%FBS）相比无统计学意义;而24h后的检测值与对照组相比P<0.01，显著高于对照组;加入20%CBP后12h和24h的BrdU检测值均显著高于对照组（P<0.05）。（2）20%CBP可明显促进脐血单个核细胞cyclin A、CDK2和CDK4的表达，但对cyclin D 1 的表达影响较小。（3）加入20%CBP继续培养12和24h后，未检测到明显的凋亡，而10%FBS组流式检测凋亡率分别为4.71%和5.76%。结论 20%CBP可通过促进造血细胞DNA的合成和细胞周期蛋白的表达，从而促进细胞增殖并显著抑制造血细胞的凋亡。
