人Ⅰ型补体受体重组腺病毒的构建和鉴定

【摘要】 目的 构建并鉴定CR ₁ 重组腺病毒，为有关基因转染研究作准备。方法 CR ₁ 目的基因经酶切插入pAxCAwt载体，构建成CR ₁ -pAxCAwt重组粘粒。磷酸钙共沉淀法转染293细胞获取重组腺病毒。结果 CR ₁ -pAxCAwt重组粘粒插入方向正确，所获腺病毒为复制缺陷型sCR ₁ 重组腺病毒。结论 采用的粘粒构建、转染方法可行，所获重组腺病毒可靠。

【Abstract】 Objective To construct the replication-deficient recombinant adenovirus coding sCR ₁ gene before the studies of gene transfection to cardiac muscle of transplanted heart.Methods The sCR ₁ -pAxCAwt was constructed by inserted the cDNA of interest into the SwaI site of pAxCAwt.Results The right direction of the insert was confirmed by restriction.Conclusion The direction of the insert must be confirmed by restriction analysis，and the DNA package protein is much helpful during the transfection of the recombinant cosmid to E.coli.

1 材料与方法

1.1 材料

1.1.2 菌株、质粒和粘粒 PUC119质粒、大肠杆菌TG ₁ 菌株由复旦大学遗传学研究所提供。粘粒载体pAxCAwt及菌株LE392为日本TaKaRa公司腺病毒表达载体试剂盒所带。
